Sorry, but the ability to map one input to many controls is currently not possible on DeSmuME Mac since the input handler doesn't support it. Actually, DeSmuME Mac has the opposite feature: You can map many inputs to one control. This is by design, as a user may have several input devices (such as a keyboard, mouse, gamepad, etc.) that they want to use. With the many-inputs-to-one-control feature, the user doesn't have to remap when they switch devices. I know that this feature is insanely useful for laptop users.
I personally don't see any practical benefit of one-input-to-many-controls, since most users prefer having precise control over their games. If a user tries to map an input that is already mapped, then the previous mapping will be removed. This is not a mistake -- this is by design. In fact, the inclusion of the many-inputs-to-one-control feature, the exclusion of the one-input-to-many-controls, and the removal of previous input mappings is the design choice used by the vast majority of mainstream apps and games.
